In Vitro Evaluation of Microhardness of Hydrophilic Pit and Fissure Sealants Reinforced with Green-synthesized Silver Nanoparticles

Abstract

Aim: This in vitro study aimed to compare the microhardness of hydrophilic pit and fissure sealants containing green-synthesized silver nanoparticles with hydrophilic sealants and conventional resin sealants on permanent molars. Materials and methods: Silver nanoparticles were green-synthesized using beetroot extract and incorporated into the hydrophilic UltraSeal XT hydro sealant. A total of 33 molars that were extracted were randomly distributed equally to three distinct groups—group I: conventionally used Clinpro sealant, group II: hydrophilic UltraSeal XT Hydro, and group III: UltraSeal XT hydro reinforced with silver nanoparticles, which were then subjected to surface microhardness testing utilizing digital display Vickers microhardness tester. Data were analyzed using Statistical Package for the Social Sciences (SPSS) software (IBM Corp. Released 2015. IBM SPSS Statistics for Windows, Version 23.0. Armonk, New York, United States of America: IBM Corp). Shapiro–Wilks test was used to determine the normality of the data. Kruskal–Wallis test was administered to compare the difference in mean microhardness scores of groups I, II, and III sealants. Results: The difference in microhardness scores [Vickers hardness number (VHN)] of the three groups was statistically significant, with reinforced UltraSeal XT hydro exhibiting the highest value (25.39 ± 1.21), followed by UltraSeal XT hydro (19.65 ± 1.35) and Clinpro sealant (19.62 ± 1.57). Conclusion: UltraSeal XT hydro reinforced with silver nanoparticles (group III) showed the highest microhardness values in comparison with the other groups. Clinical significance: Nanoparticles are gaining attention attributable to their superior physicochemical and biological characteristics, which in turn has facilitated the development of innovative materials with improved functions. Though silver nanoparticles have proven antimicrobial efficacy, their application in pit and fissure sealants is yet to be established beyond doubt. This study has laid the foundation for future research on sealants reinforced with silver nanoparticles.
